چکیده

Trans-synaptic interactions organize the multiple steps of synaptic development and are critical to generate fully functional neuronal circuits. While trans-synaptic primarily mediated by cell adhesion molecules (CAMs), some directly involve ionotropic glutamate receptors (iGluRs). Here, we review expanding extracellular proteome iGluRs. We discuss role these molecular networks in specifying formation excitatory inhibitory circuits input-specific recruitment iGluRs at synapses various types brain regions. also shed light on human-specific mutations affecting iGluR-mediated that may provide unique features human contribute its susceptibility neurodevelopmental disorders. Together, data support a view which iGluR function goes far beyond fast transmission shaping wiring properties neural

Neurosteroid modulation of ionotropic glutamate receptors and excitatory synaptic transmission.

Ionotropic glutamate receptors function can be affected by neurosteroids, both positively and negatively. N-methyl-D-aspartate (NMDA) receptor responses to exogenously applied glutamate are potentiated or inhibited (depending on the receptor subunit composition) by pregnenolone sulphate (PS) and inhibited by pregnanolone sulphate (3alpha5betaS). While PS effect is most pronounced when its appli...

Ionotropic Glutamate Receptors and their Role in Neurological Diseases

Glutamate is extensively and relatively uniformly distributed in the central nervous system (CNS) and its effects mediated by two distinct groups of receptors including Ionotropic and metabotropic glutamate receptors. Concentration of glutamate in the nervous system is much higher than in other tissues. Glutamate receptors play an important role in synaptic transmission, neural plasticity and n...

Ligands for ionotropic glutamate receptors.

Marine-derived small molecules and peptides have played a central role in elaborating pharmacological specificities and neuronal functions of mammalian ionotropic glutamate receptors (iGluRs), the primary mediators of excitatory synaptic transmission in the central nervous system (CNS). As well, the pathological sequelae elicited by one class of compounds (the kainoids) constitute a widely-used...

Synaptic NR2A- but not NR2B-Containing NMDA Receptors Increase with Blockade of Ionotropic Glutamate Receptors

NMDA receptors (NMDAR) are key molecules involved in physiological and pathophysiological brain processes such as plasticity and excitotoxicity. Neuronal activity regulates NMDA receptor levels in the cell membrane. However, little is known on which time scale this regulation occurs and whether the two main diheteromeric NMDA receptor subtypes in forebrain, NR1/NR2A and NR1/NR2B, are regulated ...

Functional Architecture of Olfactory Ionotropic Glutamate Receptors

VIDEO ABSTRACT Ionotropic glutamate receptors (iGluRs) are ligand-gated ion channels that mediate chemical communication between neurons at synapses. A variant iGluR subfamily, the Ionotropic Receptors (IRs), was recently proposed to detect environmental volatile chemicals in olfactory cilia. Here, we elucidate how these peripheral chemosensors have evolved mechanistically from their iGluR ance...
